P-69: Survival Rate Study of GnRH Potential in Inhibition of Side Effect of Drugs used in Chemotherapy

Background: Infertility problem affects 20-30% of young couples. One of the known causes of spermatogenesis disorder is chemotherapy in patients with cancer which its side effect may last up to the end of the life. The aim of the present study is to investigate the effect of GnRH on viability of ovarian follicles defect produced by anticancer drug. Materials and Methods: In the present study adult male mice aging 6-8 weeks were used. Mice were divided into 3 groups as:control, cyclophosphamid (cyclo group) and cyclophosphamid + cetrorelix(cyclo + cetro group). Cyclophosphamid was injected as IP single dose (50 mg/kg) for 5 days. In cyclo + cetro group cetrorelix injection was started one week before cyclophosphamid treatment and continued for 3 more weeks. The survival rates of the mechanically isolated follicles of groups were determined using trypan blue staining. The follicles were scored as surviving or degenerated: degenerated follicles stained blue, and surviving ones were not stained. Results: The survival rates of isolated follicles were significantly lower in cyclophosphamid groups (p<0.05). However, there was no significant difference between of control and cyclo +cetro groups (p>0.05). Conclusion: According to the results it is concluded that GnRH antagonist administration before cancer treatment could prevent the infertility side effect of vinblastin anticancer drug.
